It was analyzed 27 strains of the nodule bacteria of soybean, lucerne and galega and selected the recipient strains having the sensitivity to canamicin (25-50 мкг/мл) and the resistance to streptomicin (600-1000 мкг/мл). It was selected two strains from the fifth strains of Rhizobium galegae revealing the resistance to 50-75 mkg/ml canamicin and to 300 mkg/ml streptomicin. The adaptation of microbe cultures of R. galegae to high concentrations of streptomycin has been done. It was established that two strains (441 і 425а) of S. meliloti were the most suitable recipients for mobilizing donor E. coli S17-1(pSUP2021::Tn5) during the process of conjugation. The strains of E. coli S17-1(pSUP5011::Tn5mob) was used for mutagenesis in B. japonicam 646. As a result of Tn5 mutagenesis of S. meliloti it was obtained 200 and 120 mutant clones of nodule bacteria of lucerne respectively having the resistance to canamicin and 300 Tn5 mutants of soybean rhizobia. The frequency of occurrence of Kmr cells depended on nodule bacteria genotype used in Tn5 mutagenesis мутагенезу and had rate 1,8o10-6, 1,2o10-7 and 2,0o10-7 for S. meliloti 425a, S. meliloti 441 and B. japonicum 646 respectively.
