Karazin popup.description2 The object of study - the mechanisms of aging. The study was performed on bone marrow stem cells of male Wistar rats aged 3 months and weighing 150-200 g, were in standard vivarium conditions. Bone marrow cells were isolated from diaphysis that was washed cooled to 4 ° C sodium phosphate buffer (pH 7.4) under pressure. Cultivation was carried out for 4 days without changing the culture medium. In all experiments, initial concentration of bone marrow cells was 2,3 ± 0,2h106 cells / mL animals. Purpose - to study the role of stem cells in aging mechanisms of organisms. Methods - obtaining and culturing bone marrow cells, determining the stability of bone marrow cells obtained from young and old animals in culture and in vivo to in vitro exposure to toxic compounds. The intensity of synthesis of DNA and RNA to determine the model of partial hepatectomy impact on stem cell regeneration. In assessing the impact of stem cells on the removal of fibrosis determined activity of glutathione peroxidase, glutathione-S-transferase, lipid hydroperoxides content of total lipids and nucleic acid content, free proline, copper. Bone marrow stem cells of old animals do not yield youth for ability to proliferate in culture. Introduction toxic concentrations of copper sulfate does not inhibit proliferation of stem cells in vitro, and enhances it with more in old animals, indicating manifestation of reaction response at the bone marrow and old animals are not inferior to young animals. Culture of bone marrow cells obtained in old animals, has a higher resistance to the introduction of copper sulphate in toxic concentrations compared to cells of young animals. In the culture of bone marrow cells obtained from older animals, mostly manifested hormesis effect to copper ions compared with young animals. The ability to proliferate in old animals on the background of the development of liver fibrosis was higher than controls and higher in young animals with liver fibrosis. The introduction of the experimental animal embryonic stem cells (ESCs) significantly increased the number of hepatocytes entering proliferation in both the first and the second wave of proliferation in animals with high output proliferative activity. The introduction of ESC animals with liver fibrosis ensured restoration of altered functional activity of fibrotic liver. We can assume that the impact of ESCs on functional activity of biological systems determined not only by the properties of embryonic cells, but the biological characteristics of the object and age had no effect on these parameters.
