The object of investigation: officinal trivalent virus vaccine for the prevention of influenza and experimental samples of the new virus liposome vaccine. The aim of the study: To develop new approaches to the development of viral vaccines, create multiple vaccine preparations candidate to assess their immunobiological properties. Methods: biochemical, molecular genetic, physical, statistical, immunological. Here is given an exhaustive review of the scientific literature of the world in recent years, devoted to ways of liposomes creating and their structural and functional features and immunological activity, depending on the composition and methods of preparation. In this study we extensively investigate the protein composition of officinal viral influenza vaccines. It is proved that the total amount of protein and the number of protein components in trivalent subunit vaccine "Influvac" and virosomal vaccine "Infleksal" are less then such in split vaccine "Vaxigrip". Also the characteristics of the surface topography of the liposomes in the composition of officinal virosomal influenza vaccine Infleksal and a new experimental liposomal influenza vaccine was studied by atomic force microscopy with making a comparison. It was determined that the vesicles experimental samples surface is haracterized by absence of folding and protrusions that indicates a greater rigidity and strength of the candidate virosomal influenza vaccine liposome membrane. Based on the high immunogenicity the best sample was selected among the candidates for the new experimental viral vaccines with different physical properties of the liposomal carrier. Additionally a number of approaches were used to enhance the immunogenicity of selected liposomal sample by adding to the structure a liposome adjuvant - chlorophyllipt and aethonium and by acylation antigenic component.Also the efficacy of these methods for modifying liposomal virus vaccines in terms of antigenic activity (immunogenicity) after single and double immunization of mice in the experiment was confirmed. It is proved that, along with the achievement of high antigenic activity of these approaches allow the modification to maintain safety and low reactogenicity liposomal vaccine virus by their application.
