The aim of investigations: The development of the new bioanalytical methods for quantitative analysis of some biomarkers important for clinical diagnostics, sports medicine (for example, L- and D-lactate). Methods: microbiologicals, biochemicals, physicals, chemicals, enzymatics, biosensorics. The results of the work: - the preparative isolation and purification of flavocytochrome b2 from the cells of recombinant strain-overproducer of methylotrophic yeast Ogataea polymorpha (Hansenula polymorpha) was made. The enzyme preparations were immobilized onto different types of nanoparticles and processing protocols of noninvasive enzymatic methods for the analysis of L-lactate in human biological fluids (sweat, saliva) was done; - purified enzyme preparations was immobilized in nanoparticles of noble metals and ferromagnetic nanomaterials posessing catalytic activity; the synthesized nanomaterials was characterized by atomic force microscopy and light-FTIR spectroscopy; - it was shown that immobilized enzyme preparations of flavotsytohrome b2 onto ferromagnetic particles based on iron oxides Fe3O4 and chemically synthesited Fe0 can be used in enzymatic L-lactate analysis of reusable enzyme after its separation from the reaction mixture by magnetic separation; - it was prepared a protocol for using of flavotsytohrome b2 for noninvasive enzymatic method monitoring of lactate in human biological fluids (sweat, saliva) and biosensor analysis by directly in contact bioelectrod with the skin or oral cavity. During 2013 year it was published 39 scientific works, including 12 articles (which 8 are International), 3 manuals, 1 patent of Ukraine for invention, and 23 abstracts.
