Purpose: to obtain sequences of the mitochondrial gene COI, create a database of genes of benthic invertebrates that were collected during the seasonal biological research at the Ukrainian Antarctic Station Academician Vernadsky for further study of their evolutionary history and origin of their resistance to extreme factors. Temperature programmes were developed for polymerase chain reaction (PCR) to amplify mitochondrial and nuclear genes. Considerable variability of annealing temperatures was found for different taxonomic groups of organisms. Different capacity for amplification of nuclear and mitochondrial molecular markers was found. It was suggested a hypothesis of a greater PCR capacity of conserved sequences compared to the variable. Sequences of mitochondrial and nuclear genes of Antarctic organisms were obtained. A matrix of molecular data was compiled for phylogenetic analysis and transmission to GenBank.
