The purpose of the research was to develop the domestic diagnostic test systems (erythrocyte diagnostic kit for the IHA and the ELISA test system) for metapneumovirus infection (MPV) or poultry infectious rhinotracheitis (IRT). Methods of the research were epizootological, clinical, post-mortem, virologic, serologic, PCR diagnostics, statistical. Metapneumovirus infection (MPVI or ІRT) was established in poultry farms in 4 regions of Ukraine. The pathogen was isolated, its molecular biological properties were studied. The IHA test system was developed for the diagnosis of infection metapneumovirus (MPVI). Тhe metapneumovirus strains PVT-09 / B and PVBr-11 were adapted to the cell culture Vero. The viruses with the infectious titer of 5,72 lg TCD 50 / cm3 and 4,33 lg TCD 50 / cm3 respectively were obtained. The purification and concentration of viruses by staged ultracentrifugation for the purpose of their use in immunization of chickens and development of ELISA and IHA-diagnostics were conducted. The hyperimmune serum in chickens with titers of antibodies 2808 (IDEXX, USA) for ELISA-dіagnosticum were derived. Basic technological parameters were developed for the ELISA test system in the same serum dilution. Commission trials of IHA-test diagnostic kit for the detection of specific antibodies to the MPV were conducted. The scientific documentation for "Set of components for the diagnosis of metapneumovirus infection in IHA" (Expert excretory "Dossier" №2 from 01.29.2014 y.) was approved in SSCIBSM (Kiev). The patent №85430 was obtained. Production inspections in the poultry farms of the western regions of Ukraine proved the possibility of using IHA diagnostics for MPVI control. In the depositary of SSCIBSM (Kiev) metapneumovirus strain PVT-09/B (turkey rhinotracheitis, TRT), isolated from diseased turkeys (certificate number 590 from 11.28.2013) was deposited. In intact chickens the diagnostic serums to strains of PVT-09 / B-11, the causative agent of metapneumovirus infection of turkeys and broiler chickens according to our scheme (patent № 72060, 2012 y.) were obtained, their antigenic relationships were study. We worked out the optimum ratio of the components for the manufacture of ELISA test systems. The conversion formula of antibody titers in sera of chickens for testing them in the same dilution was calculated. It was defined the threshold of negative-positive ELISA test system and its sensitivity and specificity (comparative analysis of the results of testing serum ELISA, IHA and RN); Two prototypes of ELISA dіagnosticum were made and tested in laboratory conditions. The patent №102773 from 07.07.2015 was obtained. It was developed the regulatory documentation "Instructions for preparing and controlling the ELISA test system for the detection of antibodies to metapneumovirus infection in chicken sera. Four prototypes of ELISA diagnostic kit were made in accordance with the quality parameters provided for manual control and production. The manufacturing test in "Experimental farm of SRPS" of ELISA dіagnosticum was spent to determine specific antibodies to MPV.
