The purpose was to develop the method of genotype preservation of rare and endangered populations of poultry based on the gonad transplantation. Methods of research were biotechnological, cryobiological and statistical. The method for preservation and reproduction of rare and endangered species and populations of birds on the basis of gonad transplantation, which allows receiving not less than 23.5% of the descendants with the desired genotype was worked out. The method involves ovarian cryopreservation of day-old chicks using vitrification, training hen recipients for transplantation, transplantation of the ovaries, using of mikofenolatmofetil immunosuppressant to prevent tissue rejection. Specific binding to collagen and laminin may be used for isolation of spermatogonial stem cells from total cell population of embryonic testes and the comparative gene expression of Cdh1 and Plzf genes for assessing the degree of purification of these cells. The originality of the work was to restore the primary breed of chickens using cryopreservation and transplantation. The results of the research were implemented while preserving the genotypes of rare and endangered populations of poultry in State Research Poultry Station. The scope of use is in scientific researches and in the work of selection center for the poultry industry.
