The callus cells derived from hypocotyl and root segments of 5-day old wheat seedlings were obtained onnutrient media MS with bioregulatorsof microbial origin at concentrations 20-30 µl/l in combination with 1 mg/l auxin 2,4-D. The shoots and leaf buds were formed on MS nutrient medium with bioregulators at concentrations 10-50 µl/l with 1-2 mg/l cytokinin BAP. These shoots and leaf buds were transferred on MS medium with bioregulators for their development, growth, rooting, and further verification of their resistance to nematode in greenhouse conditions. The impact of bioregulators on increase (on 75-87%) wheat resistance to the parasitic nematode Heterodera avenaewas studied in the laboratoryconditions.The difference in the degree of homology (up to 37-65%) between cytoplasmic mRNA andsi/miRNA from cells of control and experimental plants was found using Dot-blot hybridization method.In the wheatsprouts cell-freesystemofproteinsynthesis the increase of silencing activity of si/miRNA from cells of experimental wheat plants (up to 48-78%) compared to the control was shown.
