Malignant cells with overproduction of CHI3L1 (HeLa_CHI3L1) are characterized by a high variability of morphological characters than the control cells. In the CHI3L1-producing cells, actin cytoskeleton is significantly changed compared to cytoskeleton of control cells without cverproduction of this protein. Identified changes in the morphology and cytoskeleton in cells with CHI3L1 production may be closely related to malignant transformation and increased metastatic potential of the cells. Stimulation of U251 or 293 cells by CHI3L2 protein and by glycoprotein CHI3L1 as well causes the phosphorylation of ERK1/2 and AKT. It was shown for the 1st time that the addition of CHI3L2 or CHI3L1 proteins to 293cells causes the nuclear accumulation of phosphorylated ERK1/2, and activated forms of kinases are localized in the U251 cells stimulated by chitinase-like proteinsplasm, indicating to the cell specificity of the process. It is shown that CHI3L1-induced ERK1/2 phosphorylation requires the formation of clathrin-fringed vesicles, whereas for CHI3L2-induced phosphorylation need only the initial stages of endocytosis (clustering of receptor molecules and the formation of clathrin- fringed pocket).
