In this work we were used flow cytometric and molecular-biological methods, biochemical and statistical analysis methods. The aim of the study was to characterize the influence of humoral factors in the microenvironment of the proliferation, survival, metastasis potential of tumor cells in multicellular spheroids. Determination proliferative and morphological characteristics of the spheroids and monolayer cultures in various microenvironmental conditions. The objectives of the study was to generate multicellular tumor spheroids (MТS) breast cancer cells. Determine the proliferative and morphological characteristics of MТS under different conditions of the microenvironment. As a result, it was revealed that Tamoxifen in a concentration of 10 ug/ml significantly affect the adhesive fraction of tumor cells. The cells suspension the fraction did not show such a sensitivity to tamoxifen. Interferon gamma and T-lymphocytes stimulate proliferation of several tumor cell fraction in the adhesive. But this process is accompanied by a significant increase in the number of dead cells. Interferon alpha, and vice versa MSCs inhibit the proliferation of tumor cells, which takes place against the background of a slight increase in the number of dead cells. Under the influence of the T-lymphocyte IFN gamma and the number of live and dead cells in the suspension is increased significantly fraction. The opposite picture is observed when the tumor cells were incubated in the presence of interferon alpha and its source of mesenchymal stem cells. Estradiol in a concentration of 10 ug/ml of T-lymphocytes in the system of co-cultivation and interferon gamma in a concentration 103MO/ml stimulated the formation of MTS and cell survival in the suspension.
