Ovcharenko Institute of biocolloidal Chemistry NAS of Ukraine popup.description2 First was developed the methods for the synthesis of targeted (selective to cancer cells and tumors) nanocomposites (NCs) based on nanoparticles and nanocrystals of gold with adsorption layers of doxorubicin and polysaccharides of microalgae (NPs Au-PS and NCs Au-Dox-PS, Dox-inside), functionalized for improve the selectivity of their aggregation with cancer cells by calcium folate (NCs of Au-PS-FC and NCs Au-Dox-PS-FC) and for enhance the toxic effect on cancer cells - by doxorubicin and alkaloid amitozinoberamid. It is shown that under the influence of such nanocomposites (concentration of gold - 80 mcg / ml) after the first hour of contact died more than 50% of cancers of blood cells with folate-receptors line Jurkat, and for example of the similar leukemia cells line Namalwa, which have no folate receptors - for two hours or longer. The effect is dose-dependent. It is shown that the introduction into the nanocomposite structure alkaloids or doxorubicin enhances this effect, and the concentration of doxorubicin in the nanocomposite is several times lower compared to pure doxorubicin concentration needed to achieve the same toxic effect. On the basis of the methods of synthesis of gold nanocrystals developed during the previous themes were created bright new biomarkers - modified gold nanoprisms for the detection of biomolecules and cancer cells. The effect is determined by the detection of plasmon resonance at the corners of the gold nanoprisms (surface-enhanced plasmon resonance) and mirror-reflecting surface of the crystal nanoprisms. It is shown that the increase in the spectral signal from adsorbed on the surface of nano prism biopolymer reaches 103-105 fold (which leads to determination of ultra-low concentrations). Also it was shown that the registration of selectively aggregated with gold nanocrystals of certain cancers of blood cells in the background of the vast number of healthy cells can be realized even simpler method of dark-field optical microscopy.
