The aim of the study is evaluation of functional characteristics of intersectin family of scaffold proteins in mammal neurons, as well as the role of their interaction with Epstein-Barr virus LMP2A latent membrane protein, their impact in cellular invasive properties, identification of nuclear localisation of intersectins and identification of the role of mechanisms of their activity regulation by post-translational modifications. Methods used in the study: reverse transcription - polymerase chain reaction (RT-PCR), quantitative PCR in real time, cell culture and transfection, molecular cloning, expression and purification of recombinant proteins, Western blot analysis, immunoprecipitation, fluorescent laser confocal microscopy, proteolic degradation of extracellular matrix. Obtained results significantly extend current knowledge of the role of ITSNs in formation of endocytic and signaling protein complexes in cells and can be valuable for development of new approaches for therapy and diagnostics of oncological and neurodegenerative diseases.
