The frequency of occurrences in the strain 159 of R.galegae was 4,4·10-8 and 1,6· 10-7 and in the strain 0703 of R. galegae was 1,9·10-7 and 1,3·10-7 depending on the exposure time (6 and 18 hours) when using the vector pSUP5011::Tn5mob. Transposon mutagenesis involving the vector pSUP2021 caused the frequency of occurrences 3,1?10-9 and 2,4?10-9 in case of R.galegae 159 and 1,2?10-7 and 1,8?10-7 using the strain 0703 of R. galegae. The transposon mutagenesis of R. galegae 0702 was ineffective. It was shown that the strains B16 and B20 of B.japonicum created by the vector pSUP5011::Tn5mob had poor growth and EPS contents in the growth media compared to B.japonicum T21-2 (transposon mutant with plasmid pSUP2021::Tn5). The strain of alfalfa nodule bacteria S.meliloti AC88 selected by analytical methods synthesized high amount of EPS and had high growth activity compared to S.meliloti 425а. It was established that the contents of protein in bacterial cells depended on the genotype of rhizobia. This fact can be taken into account when new express markers of symbiosis efficacy are developed.
