The aim - to study the features of gene expression of O6-Methylguanine-DNA -Methyltransferase (MGMT) reparative enzyme and its inhibiting possibility by O6-benzylguanine (О6-BzG) in patients with brain malignant gliomas to optimize the combined treatment and improve the patient state after treatment. The molecular biological and molecular genetic studies of gene MGMT expression by brain glioma cells in vitro in the presence of chemotherapy with alkylating mechanism of action by the methods of Western blot analysis and polymerase chain reaction (PCR) were conducted. The influence of reparative enzyme O6-MGMT O6-benzylguanine inhibitor on the effect of alkylating agents under short-term (72 hour) culturing the cells of human brain malignant gliomas was studied. The catamnestic data for effective combined treatment of patients with malignant gliomas based on the results of monitoring the expression level of reparative enzyme gene MGMT in body tissues and the possibility of inhibiting its activity were analysed.
