We have found that BMP preparations generated as a result of cotransfection of 85% of optimized BMP2 and 15% BMP7 plasmids are most efficient inducers of early osteogenesis of mesenchymal precursor cells. Freezing-thawing of BMP2/7 preparations does not reduce their osteoinductive properties. Polyuretan osteoplastic materials were generated as a cooperative effort with Gdansk Polytechnical Univarsity, Gdansk, Poland. These matrices are non-toxic and do not inhibit osteoblast differentiation of mesenchymal precursor cells. Our data will enable a preliminary evaluation of most effective variants of recombinant BMP preparations, primarily BMP2/7 heretodimers, to be used for enhancement of osteoinductive properties of polyuretan-based osteoplastic matrices. Such data will be an important contribution to both deeper understanding of basic molecular mechanisms in BMP signalling and practical use in translational biomedicine amd medical practice.
