Aim of investigation is the experimental study of the effect of gold nanoparticles (20 nm) and new nanocomposite systems that are based on the polypeptide EMAP II and polymer materials, on the organs of the reproductive system of male rats and on human hormone-dependent PCa tissue. Methods: general biological, biochemical, hormonal, morphological, statistical. Results: It revealed a reduction of epithelial-stromal ratio PCa xenografts transplanted to mice were injected subcutaneously with polydisperse (with an average size 26,4 nm) of the nano-Au solution in doses of 0.64 mkg/kg, 1,7 mkg/kg and 6,4 mkg/kg, but a dose-dependent effect was not observed. A significant growth inhibition of LNCaP cell cultures adding polydisperse nano-Au solution in a concentration of 10 mkg/ml, and remainance of the cell number at the control levels at the same concentration under the stimulation of proliferation of cells by dihydrotestosterone were observed. Mass cell destruction and apparent violations of their structure was not observed. Monodisperse solution (20nm) did not affect the growth of the culture. In sexually mature male rats polydisperse solution of nano-Au (5 mg/kg bw, 7 days) caused a significant reduction in weight of the seminal vesicles only. Histological examination of tissues of the reproductive system had showed inflammation and initial manifestations of atrophic changes in the ventral prostate (VP) and the absence of morphological changes in the testes of rats in experimental group. Monodisperse solution of nano-Au (0,3 mg/kg bw, 14 days) reduced the weight of the VP only, where degenerative and atrophic changes of acinar epithelium and significant inflammatory response in stroma were found. In this course, lipid peroxidation in the gland has not been changed, while in the testes nano-Au did not reduced the formation of conjugated dienes. The weight of other organs of the reproductive system and sperm content in epididimial fluide and fructose in the coagulating gland, and testosterone in blood plasma did not change under application of both solutions. Parenteral administration mono- (10 mg/kg bw) or polydisperse (1 mg/kg bw) solutions of nano-Au to castrated immature male rats for 6 days while simultaneously administered testosterone propionate (TP) in a dose of 0,2 mg/kg bw did not cause significant changes weight of accessory sex glands, as well as the structure of VP and content of nucleic acids in its tissue as compared with the effect of the TP. The effect of the new nanocomposite systems based on polypeptide EMAP II (nanocomposite - stabilized beta-cyclodextrin (CD) or hydroxypropyl-beta-cyclodextrin (HPCD) or monocomponent drug on xenograft human PCa was studied. Stable effect on the growth of xenografts was not observed. Histological studies showed an increase of inflammatory changes in xenografts of PCa when using nanocomposites EMAP II with CD or HPCD. The experimental results showed the potential anti-tumor properties of polydisperse solution of gold nanoparticles and can be taken into account in the development of new means of drug therapy for PCa.
