Kavetsky Institute of Experimental Pathology, Oncology and Radiobiology popup.description2 An influence of cytostatic agents of different mechanisms of action (cisplatin, doxorubicin, methotrexate, vincristine) on viability of cultured human tumor cells has been studied in cell lines MCF-7 (breast adenocarcinoma), А431 (epidermoid carcinoma) and А549 (non small cell lung adenocarcinoma). It has been shown that exogenous recombinant hBD-3 enhances cytotoxic effect of antimitotic agents docetaxel and vincristine, but has no effect of the sensitivity of the cells to methotrexate and doxorubicin. Expression of mRNAs of beta- and alpha-defensins hBD-1-4 and HNP1-3 was studied in MCF-7, А431 and А549 cells cultured without cytostatic agents or in the presence of cisplatin, doxorubicine, methotrexate, vincristine at the concentrations of ІС10 and ІС20. It has been shown that А431 and А549 cells express only hBD-1 mRNA, and the cytostatics have no influence on the defensin expression profile, while MCF-7 cells express mRNAs of hBD-1, hBD-2 and HNP-1-3, and expression of alpha-defensins is decreased upon the action of methotrexate Product Description popup.authors Н.В.Глушко О.С.Зубенко Погрібний П.В.
