Subject of study: mesenchymal stem cells (MSC) from human umbilical cord. The purpose of this study was to explore intercellular interactions, and to find the ways of their utilization as potential therapeutic technologies. The molecular biology techniques and following devices were used for the study: Galaxy® 170R / 170S CO2 Incubator (New Brunswick), "Tercik" thermal cycler (DNA-technology), RealTime CFX 96 (BioRad), NanoDrop1000(ThermoScientific), inverted microscope (Leica), FACS Aria (Becton Dickinson), laser scanning microscope LSM 510 META (Zeiss), and etc.. The polymer based on crosslinked modified heparin and functionalized scaffold from type I collagen were developed for the local intratissue administration of human interleukin-10 and stromal-derived factor-1a. In studies performed on the model of chronic kidney ischemia in rabbits, it was demonstrated that administration of developed polymeric carrier loaded with recombinant interleukin-10 had prominent anti-ischemic effect. The possibility to use another method for the delivery therapeutic agents, namely the encapsulation of cells in alginate microcapsules, had been also studied both in vitro and in vivo. The efficiency of such delivery method was demonstrated in vivo by amelioration of liver damage after administration of encapsulated MSC. The system for non-viral gene delivery into MSC from human umbilical cord was optimized. The MCS with enhanced expression of interleukin-10 were obtained by the optimized technique. The level of the expression by transiently transfected cells was 100 times higher than in unmodified MSC. Positive therapeutic effects of the MSC transplantations had been demonstrated in several experimental disease models (liver cirrhosis, atrophic rhinitis, experimental allergic encephalomyelitis, systemic lesions in mice, sterile inflammation). The ways for the enhancement of the observed effects were also determined. The recruitment of allogenic murine bone marrow MSC into functionalized scaffold loaded with human SDF-1a had been demonstrated. The interaction of transplanted MSC with the resident cells had been studied with the use of collagen 3D scaffolds. It had been shown that the human MSC from umbilical cord were able to mobilize resident cells, mostly represented by murine MSC. The experimental data obtained are mostly new and aimed at creating of the basis for further development of the protocols for application of MSC from human umbilical cord in clinical practice, and for the development of new technologies for the complex use of cytokines and multipotent cells.
