New methodical approaches for estimating the dynamics of accumulation of glutamate and stimulated depolarization of the plasma membrane of transporter-dependent release of glutamate from the nerve terminals of the brain using biosensor have been developed. The presence of the base signal in the preparation of the isolated nerve terminals of the brain in the absence of exogenous glutamate is shown. A methodical approach is developed for estimating the dynamics of tonic release of glutamate from nerve terminals, which is significantly increased under conditions of hypoxia and cholesterol deficiency in nerve terminals and causes the development of neurotoxicity under these conditions. The distribution of endogenous glutamate between the extracellular space and the cytoplasm of the nerve terminals is determined. The development of a method for the use of a glutamate-sensitive biosensor for the analysis of active transport of glutamate in nerve terminals of the brain will allow the use of this sensor in medicine, as well as in biochemical and biotechnological research.
