Kavetsky Institute of Experimental Pathology, Oncology and Radiobiology popup.description2 The water-soluble star-like copolymers dextran-polyacrylamide in nonionic and anionic forms were synthesized and characterized. The technique of a synthesis of double hybrid nanosystems containing nanoparticles of gold synthesized in-situ in a branched water-soluble nonionic and anionic copolymer matrix of dextran-polyacrylamide was developed and optimized. It is shown that the synthesized nanocomposites are stable in time and have a size of gold nanoparticles of 10-20 nm. A method for the synthesis of a triple nanosystem containing gold nanoparticles and a photosensitizer in a polymer matrix has been developed and its physicochemical and photophysical properties have been investigated. The analysis of the cytotoxic activity of hybrid nanosystems in relation to malignantly transformed cells of different histogenesis showed that the most sensitive among the studied cell cultures is the line of MT-4. IC30 and IC50 for these cells were 25 ?g / ml and 50 ?g / ml, respectively. At the same time, similar rates for the culture of MCF-7 cells were almost twice that of 50 ?g / ml and 126 ?g / ml, respectively. The less sensitivity to the toxic effects of nanosystems is detected in human breast cancer cells resistant to cytostatics (cisplatin or doxorubicin). Thus, 30% death of MCF-7/DDP cells was observed at 250 ?g / ml nanosystems and 50% at 500 ?g / ml. The same pattern was observed in culture MCF-7/Dox: IC30 - 375 ?g/ml, IC50 - 500 ?g/ml. It is proved that double and triple nanosystems, synthesized in the branched copolymer matrix, are time-stable (gold nanoparticle size is practically unchanged), nanosystem has a size suitable for use in laboratory experiments. It was established that the culture of human T-cell leukemia MT-4 was most susceptible to the cytotoxic effect of double nanosystem on the basis of ion dextran-polyacrylamide copolymers. For the culture of the mammary gland MCF-7, the nanosystem was less toxic in almost twice. The least sensitive to the cytotoxic effects of nanosystems were the cells of the MCF-7 lines, resistant to cytostatics.
