The objective is to estimate the status of G. lutea in the Ukrainian Carpathians through a complex analysis of ecological and genetic parameters of its populations. Methods: molecular-genetic (DNA isolation, DNA electrophoresis in agarose gel, amplification with the use of RAPD-, ISSR-, RGAP-, CDDP-, IRAP-, LP-markers), population-ecological (investigation of the plant ontogenesis, age and spatial structure, determination of the specimen density and origin, measurement of the population area and morphometric parameters of generative specimens for determination of vitality structure), mathematical statistics. To estimate the populations status of gentians (on the example of G. lutea), for the first time, the approach based on a complex analysis has been suggested and realized, and the component of it is the estimation of genetic structure of populations and correlation of the obtained data with the results of ecological research. The analysis of population parameters has been done and population dependence on the influence of natural and anthropogenic factors has been determined. It has been established that the disturbance of population stability is caused by the influence of biotic (shading with high shrubbery, dense grassing) and anthropogenic (grazing load, uncontrolled digging out rhizomes for the needs of alternative medicine) factors. Based on the study and correlation of ecological characteristics there have been determined three types of populations strategy that allow assessing the expediency and availability of the populations renewal. For the first time there have been chosen RAPD-, ISSR-, RGAP-, CDDP-, IRAP-primers to hold PCR with the samples of G. lutea DNA. Due to the molecular-genetic research of six G. lutea populations (Lemska mountain valley, Pozhyzhevska mountain, Sheshul-Pavlyk mountains, Hutyn-Tomnatyk mountain, Krachuneska mountain valley, Troyaska-Tataruka mountains) from the Ukrainian Carpathians, there has been assessed genetic diversity and determined genetic distances among populations. Genetic differentiation of G. lutea populations has been studied and genetic structure of this species has been established. For the first time the connection of genetic and ecological-geographic parameters of G. lutea populations has been analyzed. With such method used the investigated G. lutea populations have been referred to three groups: stable (Lemska mountain valley, Sheshul-Pavlyk mountains, Pozhyzhevska mountain), relatively stable (Hutyn-Tomnatyk mountain), and unstable (Krachuneska mountain valley, Troyaska-Tataruka mountains). The genetic peculiarities of agro-population from Pozhyzhevska mountain have been studied and the probability to use artificial planting aimed at G. lutea consevation has been estimated. For the first time on the example of G. lutea the efficient indicator of informativeness of primers has been chosen and there has been developed a system of molecular-genetic markers for estimating genetic polymorphism of G. lutea that is based on the use of different DNA areas as primers. On the ground of the studied G. lutea ecological parameters and estimated genetic diversity of its populations, the recommendations for conservation of this rare species have been worked out. With the results of ecological-coenotic and molecular-genetic research used as well as the data on the trophic needs of gentians there has been developed and tested the methodology for adaptation of grown in vitro plants to conditions ex vitro and held the repatriation of these plants into the natural conditions of the Ukrainian Carpathians. There have been developed ecological-genetic basics for Gentiana species consevation and suggested the approach to classify gentian populations according to the degree of their stability. The suggested method can be used for estimating the status and renewal of other species populations of Gentiana genus and affined to gentians mountainous species. The results of complex estimation of populations status allow to forecast populations disturbance at the initial stages and promote biodiversity conservation that is in the mainstream of scientific research defined by the international program document "The Convention on Biological Diversity" and the Presidium of NAS of Ukraine.
