Changes of oxidative homeostasis and state of cardiomyocytes in case of type 1 diabetes mellitus (DM1) and in presence of pharmacological correction. The aim of the research. The study of role of oxidative stress (OS) in case of DM1, early manifestation processes of experimental diabetic cardiomyopathy (DC) in rats and their changes under the influence of N-acetylcysteine and losartan. The methods of the research – clinical, instrumental, biochemical, electronomicroscopical, statistical. 110 Wistar rats (220–250 g) were investigated. Also, 36 children (20 boys and 16 girls) aged from 11 till 17 years old were studied. In case of DM1 in patients as well as in experimental studies were indicated decreasing of antioxidant activity (AOA), intensification of OS and lipoperoxidation. After 1 month of experimental DM1 in rats were observed decreasing of cytoprotective autophagy of cardiomyocytes and electrocardiographic changes. The latest ones were observed on the 2nd week of experiment. The relationship structure- antioxidant activity of N-acetylcysteine and losartan in vitro and in silico was studied. The cardioprotective action of N-acetylcysteine and losartan and their combination were detected. N-acetylcysteine lowered hyperglycemia-induced DC’s manifestation by its antioxidant action. N-acetylcysteine provided membranoprotection, decreasing intensification of OS and lipoperoxidation, restored autophagic cytoprotective activity, heart rhythm, and ventricular contractile activity of myocardium that can become a new therapeutic strategy for patients with DC.
