The interaction of nanocrystals with the cell plasma membrane leads to its damage. In addition, intracellular membrane vezicles with DNA were observed in the sites of interaction with nanoparticles. Incubation of isolated granulocytes with nanocrystals resulted in decondensation and externalization of chromatin. Nanodiamonds trigger granulocyte activation and NET (neutrophil extracellular traps) formation. Based on our data, we proposed a mechanism of interaction of nanocrystals with the cell membrane. The particles whose size is comparable to the thickness of lipid bilayers, penetrate through the plasma membrane and disturb its barrier function. Inside the cell, the damaged membranes fuse with primary lysosomes causing lysosomal leakage and triggering a cascade of events involving ROS production and chromatin externalization. Inosine released from dying or dead cells stimulates cell proliferation via adenosine receptors. Inflammatory reactions and tissue damage triggered by the small particles (10 nm) which acted like adjuvants of specific humoral and cellular immune responses.
