Research report: p.49., 8 Fig., 3 table, 54 sources. The object of research - tumor spheroids line MCF-7, mesenchymal stem cells of man (MSC), interferon alfa (IFNa). The purpose of the study was to investigate the effect of cellular origin factors (IFN? and factors from MSC) on the antigenic profile, clonogenic and metastatic potential of monolayer culture cells and various subpopulations of cells of a multicellular tumor spheroid (BPS). Methods of research - cultivation of monolayer cell culture and BPS, immunohistochemical, analysis of proliferative and adhesive properties of cells, statistical analysis methods. Different reactivity, viability of tumor cells in the monolayer and BPS under the influence of IFN and humoral factors from MSK were established. The change in the growth type of the tumor population causes changes in the expression of markers of the epithelial-mesenchymal transition (cytokeratins, adhesion molecule of epithelial cells and vimentin). Vimentin is expressed only during spheroid growth. IFNa and humoral factors from MSC have a cytotoxic effect on tumor cells, and also reduce their migratory ability. IFNa leads to the appearance of signs of differentiation of tumor cells. Forecasting assumptions on the development of the research object - it is expected that the results of the studies should become an indispensable stage for studying the ways and mechanisms of the action of systemic personalized therapy aimed at stem tumor cells based on BPS.
