The aim: metabolic engineering for improvement of the yeast Ogataea polymorpha for increased ethanol production during alcoholic fermentation. The aim of stage: to increase the efficiency of xylose transport by engineering of modified forms of O. polymorpha Hxt1 transporter. Methods: genetic engineering, microbiological, biochemical. Using genetic engineering approaches, O. polymorpha yeast strains were constructed with increased affinity of the Hxt1 transporter to xylose and reduced to glucose to achieve effective transport of both sugars into the cell. To increase the specific xylose uptake rate the modified Hxt1 was engineered by substitution of asparagine to alanine at position 358. Furthermore, N-terminal lysine residues of Hxt1 predicted to be the target of ubiquitination were replaced for arginine residues. The modified versions of Hxt1 was overexpressed in hxt1 mutant, unable to produce ethanol during high-temperature alcoholic fermentation.
