Aim of research – isolation and identification of Gaeumannomyces spp based on cultural and morphological characteristics and pathogenicity factor, testing of set high specific primers on DNA from these isolates for future development of molecular test-system for diagnosis take-all and Fusarium root rot of wheat. Methods of research – field (sampling of winter wheat plants and visual diagnosis of their diseases), laboratory ( isolation of fungal pathogens in pure culture, study of their cultural and morphological characteristics, screening isolates presumably identified as G.graminis), green-house experiments (artificial inoculation, pathogenicity identification and confirmation plant in vivo), molecular-genetic (design and analyze in BLAST new sets of specific primers for PCR identification species and variants Gaeumannomyces and their testing on DNA from isolates Gaeumannomyces). The importance of results is the creation and confirmation of high-level specificity of primers complementary to ribosomal claster of DNA on the isolates of G.graminis. Using these primers the amplicons of 400 bp were revealed in fungal culture, which correspond to variants of G.graminis var.tritici. The results of molecular analyzes confirm the occurrence of take-all on winter wheat in farms where the samples were taken. Since, only using molecular diagnosis methods the variant G.graminis var. tritici characterized as a highly virulent was detected on the wheat samples this is an evidence that the molecular methods of root disease of winter wheat should be introduced into practice of phytosanitary monitoring of cereals crops.
