This compound acts as a complete noncompetitive inhibitor for the plasma membrane Ca2+,Mg2+-ATPase. Calix[4]aren C-97 and C-99 at a concentration of 100 nM inhibited Са2+ accumulation in isolated mitochondria of smooth muscle and reduced the concentration of this cation in the matrix. It was shown the ability of calix[4]aren C-99 to penetrate through the smooth muscle cell plasma membrane and its influence not only on the ATPase activity of myosin, but on the integrity of the aktomyosin complex. Nitric oxide or its derivatives moderately reduce the mitochondrial transmembran potential, which is confirmed by the flow cytometry data. It is proven that antagonists of calmodulin reduce the level of polarization of myometrial mitochondria membranes.
