Goal of the work - to evaluate the effectiveness of proposed different methods MSC cryoconservation in vitro using proliferative activity and phenotype researching, cryoconserved cells influence on the moving functions and behavior reactions, spinal cord morphological changes after their intrathecal injection to the experimental animals. During last years the progress in CNS inflammatory-degenerative damage treatment is connecting with cell technology development. One of the most perspective for using in clinical practice are Warton's jelly cells (from the umbilical cord), which can be simply isolated without any moral ore ethical conflicts. Despite the perspective of clinical using human umbilical cord MSC, the question of choice of optimal MSC long-term storage (cryoconservation) method without DMSO and taking into account freezing condition, kind of cryoprotectors and terming of storage is arising. Many variants of cultured MSC cryoconserving, composition and concentration of cryoconservants was studied. The cryoconserved MSC cell vitality, their proliferative and phenotypic properties changes under cryoconserving action was researched in vitro. The obtained on the cell level results allowed to conclude on the possibility the significal reduction DMSO concentration in the freezing medium to the 4% instead 10% at the expense of no less 6% trehalose addition. The EAE modeling on the 50 rats, animal behavior reaction in the test "open field", immunology and morphology tissue and blood experimental animal samples after intrathecal injection of MSC, cryoconserved by the different method, investigations were carried out. Obtained in vivo results also confirmed the opportunity of cryoconservants concentration optimization at the MSC freezing, which reduced DMSO toxical effect to the cells and other negative side effects development.
