The goal of the work is to work out sample preparation protocols, vitrification schemes and registration of temperature schedules. Determination of the effect of vitrification on the survival, differential and proliferation potential of mouse mesenchymal stem cells (MMSC) and human blood cells. Methods of research and equipment - MTT-test, test for cell viability with 7-AAD, system for the preparation of leukocytes for cryopreservation, a mock-up sample of the device for ultrafast freezing, the freezer Arctico, photoelectric colorimeter Labsystems Multiskan MCC/340. The study of the effect of the composition of cryoprotective mixtures on the freezing rate (with ultrafast and slow freezing) on ??the survival of MMSC cells and human blood cells was performed. It has been found that when using the method of slow cooling, the most appropriate is the use of cryoprotective solutions with arabinose and maltose, which increase the number of cell survival by 34 and 45.4%, respectively, while using the method of ultrafast cooling, the most effective solutions with fructose, glucose or maltose, which increase cell survival by 49, 43.3 and 53.3%, respectively. The results obtained in the course of the research and the acquired protocols were reflected in the application of the protocols for cryopreservation of donor stem cells obtained from human peripheral blood using cytophoresis procedures.
