Kavetsky Institute of Experimental Pathology, Oncology and Radiobiology popup.description2 As a result of research in the in vitro system, for the first time, general mechanisms of exogenous LF action have been identified: cell cycle blocking in the G2 / M phase, adhesion violation, change in the ratio of protein-regulators of apoptosis, decrease in the number of stem cells, increase in the number apoptotic cells. It has been established that expression of LF in tumor cells of human breast and prostate cancers is related to their sensitivity to hormone therapy. We carried out the exploration of effects of exogenous LF administration at concentrations of 1 mg / kg and 10 mg / kg on the in vivo model system in animals with carcinosarcoma, Walker-256. We have observed suppression of tumor growth, more pronounced at a concentration of 10 mg / kg (44%), which is confirmed by the results of morphological studies: segregation of tumor cells, minor increase of their size, picnotic changes, nuclei hyperchromatosis, phenomena of necrobiosis and necrosis, and as a consequence of a decrease in the total number of tumor cells. Restructuring in the lipid composition of carcinosarcoma Walker-256 cells led to a decrease in the microviscosity of their plasma membranes. On the clinical material of patients with BC and PCa it was shown that LF, in comparison with other serum inflammatory markers (C-reactive protein, lactate dehydrogenase, lactate), is the most informative, since its levels correlate with the overwhelming majority of clinical and pathological characteristics of these diseases. Survival of patients with PCa with blood LF levels not exceeding 0.27 rd. unit., was 73% compared with the survival rate of patients (57%), who had higher level of LF. It has been established that in patients with breast cancer, the level of expression of serum miR-214, that is involved in LF regulation, is significantly higher than in patients with benign tumors and controls. It has been shown that in patients with PCa there is a relationship between the level of expression of circulating miR-214 and such clinical and pathological characteristics as the presence of metastases in the lymph nodes, the increase in serum PSA level and the degree of malignancy of tumors by Glison. For the first time as a result of comparison of LF expression in tumor cells in patients with benign and malignant tumors of the breast and prostate, a different orientation of expression of this iron binding protein has been established. A number of regularities regarding the level of LF expression in the tumors of patients with various manifestations of the clinical course of BC and PCa were revealed: the inverse correlation between the level of LF expression and the size of tumors and the presence of metastases in patients with PCa, as well as direct correlation between LF expression in tumor cells and overall survival of BC patients, which is the basis to recommend this indicator as a prognostic marker for the BC course.
