It was decided to use sequences that reproduce the fragments of the rpoB gene of M. tuberculosis, namely 1) normal, unmutated gene, 2) with mutation in codon 531, 3) with mutation in codon 526. Among the oligonucleotides having the lowest ability to undesirable intramolecular interactions, a 21-mer fragment (codons 525-531) was selected, and the corresponding three probes were named PNt, P2t and P3t.
