The purpose of the work - Evaluation of the value of the interaction of cellular and endocrine factors for the implementation of neuroprotective effect of neural stem cells in damages of the central and peripheral nervous system Models: cuprizon model demyelinization, hemiparkinsonism, injury of the sciatic nerve, administration of recombinant human proteins and hormones. Methods: Cultural, biochemical, laser flow cytometry, histological, confocal and electron microscopy, "open field" test. Used equipment: Thermo CO2 incubator, Labconco laminar box of biological safety, FACSAria "Becton Dickinson", CL30R laboratory centrifuge, Olympus IX71 microscope, Kern & Sohn electronic scales. Main results: The pathogenetic factors of neurotoxin influence on the structure and functional state of the central nervous system (CNS) neurons (T-lymphocytes, macrophages, malonic dialdehyde) have been determined. Leukemia inhibitory factor (LIF), interleukin-10, fibroblast growth factor, melatonin, thyroxin, and estradiol have a positive effect on in vivo morpho-functional CNS changes induced by cuprizone; The effect is most pronounced in LIF and melatonin and depends on the age of the mice. Influence of hormones and cytokines by mediated changes in the factors of immune and oxidative stress. Melatonin and LIF exhibit a remyelinization effect on the cuprizone demyelinization model in vitro. In rats with hemiparkinsonism, melatonin improves the structure and function of CNS neurons; in the brain, the number of active macrophages falls and the blood level of the thymulin increases. In mice, melatonin and LIF stimulate reparative regeneration of the sciatic nerve. The results of the work are presented in the form of a report, 9 articles, 6 patents, 12 theses.
