The problem is solved by the use of rigid linkers connecting molecules of fluorescent dyes, as well as by increasing the water-solubility of dyes by introducing sulfonate, phosphonate and quaternary ammonium groups, which reduce the aggregation of dyes in the formation of multifluorophore systems. The regularities concerning the occurrence of chemical reactions for obtaining of such dyes and their functionalization by homo- and heterobifunctional cross-linkers have been clarified. It has been established that connection of the dye through a short linker results in quenching fluorescence due to non-radiation resonance energy transfer, and the use of flexible linkers reduces the fluorescence of dyes due to their aggregation. The possibility of using dyes to create FRET pairs with fluorescent (R-phycoerythrin and peridinin-chlorophyll-protein complex) and non-fluorescent (BSA, HSA, HRP) proteins is shown. On the basis of synthesized compounds prototypes of new fluorescent labels and probes with increased brightness were developed, which can find practical application in biomedical research and it was developed a sensitive method for determination of the conformational changes in proteins.
