The aim: to assess the cytogenetic effects in vivo and ex vivo in lymphocytes of cancer patients following external and internal radiation treatment, depending on the irradiation volume and exposure regimen. Research methods and techniques: cytogenetic analysis of human blood cultured lymphocytes, statistical methods of data processing, microscopes Axioskop, Micros MC300X and Olympus, system ASIS. Theoretical and practical results: The peculiarities of chromosomal aberrations outcome and their distribution in human lymphocytes irradiated in therapeutic doses ex vivo with simulated local radiation exposure were shown. The level of chromosomal exchanges during ex vivo irradiation at 2 Gy was significantly higher than spontaneous and exceeded 20 % - 25 % in all patient groups. The highest level of chromosomal damage was found in patient group with lung cancer at a dose of 4 Gy. A high informativity of cytogenetic test in vivo and ex vivo and of selected experimental model for detecting local irradiation and radiosensitivity of lymphocytes in patients with different tumor localization were demonstrated. These data are of importance for correct assessing the impact of therapeutic irradiation on normal tissues at the chromosomal level. Novelty: new knowledge was obtained about the character of chromosome damage outcome due to ex vivo irradiation of lymphocytes in cancer patients with different tumor localization.
