Experimental study of the effect of X-radiation and the combined effect of X-radiation and a COX-2 inhibitor - meloxivet on the content of VEGF and PGE-2. The purpose of the work is to determine the mechanisms of participation of cyclooxygenase-2 selective inhibitors in combination with ionizing radiation in blocking tumor angiogenesis to substantiate their role in radiation therapy of cancer and to develop recommendations for new technologies of combined radiation and lung cancer of the lung. Methods of research and equipment: clinical and instrumental, immunoenzymometric, electron-microscopic, dosimetric, statistical; the immunoenzymometric analyzer Immunochem-2100, sliding microtome MC-2, the universal dosimeter UNIDOS_E, the linear accelerator Clinac-600, RUM-17. Theoretical and practical results: it has been established that when using conformal radiation therapy in combination with a COX-2 inhibitor - Ranselex, a positive effect is observed in 100% of patients with HNT and in 80% of patients with NSCLC. Partial regressions prevailed, the frequency of which increased in patients with HNT and NSCLC 1.8 and 4.5 times, respectively. The mechanisms of the influence of different doses of ionizing radiation on the level of VEGF in the serum of tumor-bearing rats were determined: at a total dose of 1 Gy, stimulation of the neoangiogenesis process is observed, and at a total dose of 10 Gy, the process is slowed down. It has been proven that the combined effect of irradiation and meloxivet reduces the levels of VEGF and PGE-2 in tumor-bearing rats. Novelty: new data were obtained on the effect of the combined action of radiation therapy with Ranselex on reducing the levels of pro-angiogenic markers VEGF, COX-2 and PGE-2 in the serum of patients with NSCLC and HNT. The scientific data on the mechanisms of influence of different doses of ionizing radiation and the combined effect of radiation and the COX-2 inhibitor on the VEGF level in the blood serum of rat tumor carriers are supplemented, which is associated with the COX-2 blocking mechanism and indicates its effect on slowing down the process of angiogenesis.
