Object of the study: water-soluble biocompatible pristine C60 fullerenes. The purpose of the work was to assess the effect of biocompatible pristine C60 fullerenes on the growth factors and extracellular matrix protein receptors in vitro and the morpho-functional state of the liver under rat hepatitis and hepatocellular carcinoma in vivo. Methods: histological, biochemical, cell culture, immunocytochemical, mathematical modeling and statistical analysis. It was shown that C60 fullerenes acting under liver acute inflammation contribute to suppressing of its morphological and biochemical manifestations and restore the liver functional state. Anti-inflammatory effects of C60 under chronic inflammation were less pronounced, in addition, the features of kidney and pancreas dysfunction occurred. C60 fullerenes were capable to suppress the fibrosis and neoplasia development in liver and contributed to maintaining its normal functional activity after prolonged action since the initiation of liver damage. The effects of C60 fullerenes on the liver during the onset of in situ cancer were manifested by reducing the liver damage score and restoring its functional activity, however, the features of hepatocytes cytolysis, as well as the negative effects of C60 on the pancreas, were preserved. Moreover, positive effects of C60 fullerenes were more pronounced compared to those of the reference - the classical cytostatic drug 5-fluorouracil. C60 fullerenes suppressed the expression of EGFR and cytokeratin receptors by HepG2 cells (liver carcinoma) in a dose-dependent manner and were capable to block the ATP binding site of EGFR and FGFR, as has been shown by mathematic modeling. The obtained data let us suggest the anti-inflammatory, anti-fibrotic and antitumor activity of C60 fullerenes, as well as their ability to downregulate the signaling pathways associated with EGFR, FGFR and cellular matrix protein receptors.
