The purpose is to find out the expression profiles of genes of antimicrobial peptides from Scots pine under the action of bacterial and fungal phytopathogens to identify the genes that are induced in response to biotic stress, to perform the structural and functional characteristics of these genes and to determine the promising use of these as molecular markers of signs of resistance to phytopathogens, as well as develop methods for molecular identification of pathogens of forest infections. Methods of research are forestry, microbiological, molecular biological, genetic engineering, bioinformatics, statistical. The differential expression of the defensin genes in pine seedling tissues in response to pathogens with different life-style was revealed. The molecular cloning of the defensin 1 gene and the lipid transfer protein gene from Scots pine have been performed and their intron-exon structure is established. Recombinant PsDef1 and PsLTP1 were obtained by expression in the bacterial system and their polyfunctional properties were found. The molecular-genetic methods of diagnostics of root rot, fusariosis and bacterial dropsy of pine trees are developed, which make it possible to determine the pathogenic factor in the early stages of the development of infection. A method has been developed to increase the resistance of seedlings to infectious lodging. The algorithm of estimation of potential stability of genotypes of Scots pine to infectious defeat is offered. Areas of use are tree physiology, forest phytopathology, ecology, forestry.
