The object of the study is the immunological mechanisms of oncogenesis and structural changes in the spleen tissue of animals with simulated adenocarcinomatosis of the colon. The purpose of the research: to find out the features of the development of metabolic, immunoreactive disorders and morphofunctional reorganization of the structural components of the spleen under conditions of experimental oncogenesis. Research methods: experimental - simulation of chemically induced carcinogenesis in outburst white rats with the development of colon adenocarcinoma in situ; morphological - study of the regularities of the structural organization of the spleen on the microscopic and submicroscopic levels; immunohistochemistry - determination of the degree of expression of the Bcl-2 gene; Immunological - determination of serum immunoglobulins A, M, G, circulating immune complexes, IL-1?, IL-6, IL-2, IL-4, IL-10, TNF? concentrations; biochemical - research of indices of endogenous intoxication, activity of factors of oxidative stress, enzymatic and glutathion-dependent parts of antioxidant system; statistical Under conditions of experimental carcinogenesis in the organism of experimental animals, there are significant metabolic disturbances that manifest themselves as increased manifestations of the syndrome of neoplastic endotoxemia; the development of oxidative stress and the imbalance of the functioning of enzymatic and glutathion-dependent units of antioxidant defense. Changes in immunological reactivity are accompanied by an increase in concentrations of Ig A, Ig M, Ig G, and a significant accumulation of circulating immune complexes. Significant changes in the cytokine matrix were observed, which was manifested by a significant increase in the concentrations of IL-4, IL-6, IL-10, TNF? with a significant decrease in the content of IL-2. Histological studies of the spleen of animals in the dynamics of the formation of chronic neoplastic intoxication, induced by the introduction of dimethylhydrazine, revealed the development of destructively degenerative changes in the spleen, stroma, red and white pulp of the spleen, the severity of which increased in accordance with the observation period. The expressed enhancement of expression of the Bcl-2 gene expression in the spleen tissue of the experimental animals was established: the number of cells expressing the Bcl-2 gene on the edge of the red pulp and periarterial lymphoid muff / marginal zone and in the most red pulp significantly increased, maintaining the intensity of the reaction (+++ ), and a moderately pronounced positive reaction (++ -) in single cells, scattered around the periphery of the entire hermetic center of the white pulp.
