The spatial, energy, and electronic characteristics of the molecules of piroxicam and meloxicam have been established using Quantum Structure Activity Relationship (QSAR) methods of quantum chemistry. The analysis of the results of experimental studies indicates that the addition of caffeine to oxicamam increased the anti-exudative activity, reduced inflammatory marker values (sialic acids and C-reactive protein), analgesic activity (central and peripheral genesis), but piroxicam - only of central genesis. Addition of caffeine to piroxicam promoted an increase in the antipyretic activity of piroxicam, normalization of the cognitive activity of rats, positively influenced the grooming, stimulation the amount of urination. Additin caffeine to meloxicam contributed to an increase in vertical motility, the amount of defecation, the normalization of cognitive activity in rats. The paper provides a quantum-chemical and experimental justification for the feasibility of creating new domestic two-component pharmaceutical compositions (piroxicam + caffeine and meloxicam + caffeine) with anti-exudative and analgesic action and can be recommended for further study.
