The aim of the work was to design and develop molecular approaches for expression study of EPF1, EPF2 and MUTE genes from drought tolerant and drought susceptible bread winter wheat cultivars. The experiments were carried out using the equipment of domestic and foreign origin at the level of high international standards applying modern methods of molecular biology and bioinformatics. The following set of works was performed: genomic specific primers were designed for amplification of complete sequences of EPF1, EPF2 and MUTE genes localized on genomes A, B and D of bread wheat. The sequences of EPF1-A1, EPF1-B1 and EPF1-D1 genes of three drought tolerant (Podolianka, Astarta, Odeska 267) and three drought susceptible (Natalka, Poliska 90, Darunok Podillia) bread winter wheat cultivars of domestic breeding were amplified and sequenced. The performed sequence analysis showed the presence of a small intra-type polymorphism for the coding sequences of the EPF1-A1 (two single nucleotide substitutions in the second exon for Podolianka and Odeska 267 cultivars) and EPF1-B1 genes (single nucleotide substitution in the second exon for Podolianka and Astarta cultivars). The high level of polymorphism was observed for the coding sequences of EPF1-D1 gene (Natalka and Astarta cultivars - seven single nucleotide substitutions in the first and second exons and two insertions in the second exon). Based on the obtained sequences, the structure of EPF1-A1 EPF1-B1 and EPF1-D1 proteins was modeled. Also, using the available EPF1 protein sequences, the phylogenetic analysis of EPF1 proteins from different plant species was conducted. In addition, during the reporting period a physiological field experiment was conducted to further simulate the drought conditions and obtain sufficient quantities of wheat plant material for subsequent studies.
