Kundiev of National Academy of Medical Sciences of Ukraine" popup.description2  The toxicological studies on in vitro models (cells culture of different origin) revealed patterns and features of the toxic action of nanoparticles (NPs) of metal compounds (CdS, PbS, Fe2O3): cell membrane damage, respiratory disturbance of mitochondria, protein synthesis suppression, stimulation of oxidative stress and DNA damage. It has been established that the severity of cytotoxic and genotoxic effects is determined by the size of the particles, their concentration, the metal-base toxicity and cell sensitivity. The target organs of the unfavorable influence of metal NPs can be the liver and kidneys. It was found that NPs CdS (4-6 nm) and PbS (12.5 nm) had a higher cumulative capacity, which contributed to their accumulation in rat organs (liver, spleen, heart). The peculiarities and severity of the toxic effects of metal NPs, their hemato-, hepato- and nephrotoxic effects depends on the size, dose and exposure time. Hematotoxic activity was characterized by a shift in the process of heme synthesis and hematopoiesis, hepatotoxicity - by changes in metabolic processes, indicating toxic damage to the liver. NPs of CdS and PbS exhibited immunotoxic effects, while Fe2O3 stimulated the natural resistance of the organism. Metal NPs stimulated oxidative stress and suppressed the antioxidant defense system. According to the results of the conducted studies, the level of potential hazard of CdS and PbS NPs is characterized as high, but Fe2O3 – as medium. Positive effects have been established (reduction of metal accumulation in the body, improvement of integral, hematological and biochemical parameters) with the use of medicines with membrane protective, antioxidant, hepatotoxic and neuroprotective properties (Quercetin - by the action of Fe2O3; Tiocetam - CdS and PbS; Tiocacid 600 T (α-lipoic acid) - under the influence of PbS) can recommend them for the prevention of toxic effects of nano- and microparticles of heavy metals (cadmium, lead, iron).
