Aim: to study the survival, viability and functional activity of cord blood nucleated cells, including hematopoietic progenitor CD34+ ones, as well as the possibility of preventing/inhibiting the development of apoptosis in them during cryopreservation in the solutions containing DMSO and substances with antioxidant properties. Modern research methods were used in this study such as cytometry and spectrophotometric methods. As a result of the study it has been found that using of cryoprotective mixtures containing DMSO at a concentration of 7.5 and 10% and glutathione at a concentration of 1 and 3 mM for cryopreservation of cord blood nucleated cells, including hematopoietic progenitor ones, provides an increase in the amount of survived and viable cells after freeze-thawing, and allows preserving in a viable state up to 80% of nucleated and up to 85% of hematopoietic progenitor cells. Cryopreservation of nucleated cells under the protection of DMSO (7.5 and 10%) and glutathione in efficient concentrations (1 or 3 mmol) has been shown to allow preserving a greater amount of survived and viable nucleated cells after transfer to the conditions, modelling physiological ones as compared with similar conditions without antioxidants. Using of glutathione during cord blood nucleated cells’ cryopreservation has revealed the possibility to reduce the efficient concentration of DMSO down to 5%, which ensures a decrease in its toxic effect on cells, and consequently the achievement of survival and viability indices at the same level of the samples cryopreserved with 7.5 and 10% DMSO no antioxidant use.
