Information × Registration Number 0219U102245, 0116U004864 , R & D reports Title The application of metabolic engineering approaches for construction of Saccharomyces cerevisiae strains able to overproduction of glycerol under anaerobic conditions popup.stage_title Head Dmytruk Kostiantyn V., Registration Date 23-12-2019 Organization Institute of Cell Biology popup.description2 All previously applied genetic modifications (attenuation of TPI1 gene expression (encoding triosophosphatisomerase); overexpression of fused GPD1-GPP2fus gene (encoding glycerol-3-phosphate dehydrogenase and glycerol-3-phosphatase); expression of the truncated form of ILV2 gene (encoding acetolactate synthase) and the truncated form of the FPS1m gene (encoding a modified glycerol membrane transporter) under the control of a strong constitutive ADH1 promoter) were combined in the genome of one recombinant S. cerevisiae strain BY/tpi25/gpd1gpp2f /fps1m/ilv2. The resulting strain produced 9 times more glycerol under microaerobic conditions and 4.7 more glycerol under anaerobic conditions compared with the wild type strain BY4742. The BY/tpi25/gpd1gpp2f/fps1m/ilv2 strain was used for further selection of resistant mutants on medium with high salt concentration (2 M KCl), however, such selection did not provide further increase in glycerol production.
