During the reporting period, the redox and energy state of MCF-7 mammary adenocarcinoma cells was studied under conditions of monolayer and spheroid growth, as well as in the enrichment of multicellular tumor spheroids (BPS) cultures with stem tumor cells (SPC). Object of research: MCF-7 mammary adenocarcinoma cells. Research methods: cultivation of an adhesive and spheroid cell culture, spectrophotometric and statistical methods. The aim of the study was to compare the redox and energy state of MCF-7 mammary adenocarcinoma cells under monolayer and spheroid growth, as well as in the enrichment of BPS SPK (zBPS) culture. Objectives of the study: to compare the content and activity of enzymes of the antioxidant system of cells: MDA, KGB, HB, SOD, CAT, GP, GT, as well as energy metabolism enzymes: G6FDG and LDH in 2D culture, BPS and zBPS. As a result of the work, it was revealed that the phenomena of oxidative stress under the conditions of MCF-7 cell growth in the form of BPS are enhanced in comparison with monolayer growth and approach the status in tumor in vivo. The processes of peroxidation and neutralization of free radicals in BPS, provided they are enriched in SPC, are suppressed in comparison with unenriched BPS. It was also shown the intensification of the reactions of the pentose phosphate shunt in BPS enriched with SPC, and the inhibition of glycolysis reactions, in comparison with the cells of the monolayer culture and BPS that were cultured under ordinary conditions. Scientific and practical significance. It is expected that the use of zBPS in the study of SPC can be used to create and test systemic antitumor therapy aimed at SPC.
