The purpose of the work is to determine the effect of the vitrification method on the activity of enzymes of the redox system of mouse mesenchymal stem cells in multicellular organoids. Research methods and equipment - UV spectroscopic methods for studying the activity of enzymes, a prototype of a device for ultrafast freezing, an Arctico freezer, a Labsystems Multiskan MCC / 340 photoelectric colorimeter. A study was made of the effect of the composition of cryoprotective mixtures on the state of enzymes of the redox system of MCF-7 cells during fast and slow freezing. It was found that when using the fast freezing method, the functioning of enzymes involved in redox processes is significantly less disturbed than when using the slow freezing method. It is established that when using the method of rapid freezing, it is most appropriate to use a cryo mixtures based on solutions of galactose, lactose, and maltose. In cryo mixtures with these sugars, the highest activity of the studied enzymes and a low concentration of malondialdehyde were observed, which indicates a minimal violation of enzymatic redox processes in the cells after thawing. When using the slow freezing method, cryo mixtures containing xylose and galactose are effective. The most effective for slow freezing was xylose-based cryo mixturе.
