Bogatsky, Physico-Chemical Institute of the National Academy of Sciences of Ukraine popup.description2 Object of study - free and immobilized in poly-N-vinylpyrrolidone tyrosinase. The purpose of the work is the improving of the method of isolation and immobilization of Agaricus bisporus mushroom tyrosinase to develop reusable economic biocatalysts suitable for the production of biologically active compounds. 20 derivatives of benzylideneaniline and related compounds were synthesized with 65-88 % yields. It was shown that 3 of investigated compounds (3,4-dimethoxybenzylidene-2-aminophenol, 2,3,4-trimethoxybenzylidene-2-aminophenol, 3,4,5-trimethoxybenzylidene-2-aminophenol) are effective tyrosinase inhibitors (IC50 4-4.8 times lower than kojic acid). Biocatalyst for the synthesis of L-DOPA based on tyrosinase immobilized in poly-N-vinylpyrrolidone has been developed. It was revealed that in aqueous-organic medium the activity of the immobilized preparation exceeded the activity of free tyrosinase by 30%. Immobilized tyrosinase retains high phenoloxidase activity for 6 cycles of use.
