The purpose of the work was to determine the relationship between the structure and genes functioning system which causes the stomata formation, and the physiological and biochemical characteristics of Ukrainian winter wheat cultivars in conditions of insufficient humidity. Research methods included bioinformatic approaches of nucleotide and amino acid sequences study, molecular biological analysis (molecular cloning and sequencing, PCR, real-time PCR), physiological and biochemical analysis of the level of wheat drought tolerance. EPF/MUTE gene structures for six bread wheat cultivars were investigated; changes in physiological and biochemical parameters have been determined and stomata on the abaxial and adaxial sides of wheat leaf have been characterized; genes expression patterns were established; the correlation between the polymorphism and the physiological characteristics of the varieties was studied. As a result, EPF/MUTE sequences from different subgenomes of 6 varieties have been characterized. The differential expression of EPF/MUTE genes was observed only on the early stages of leaf development. Comparison of expression profiles supported the hypothesis of a possible role of EPF1 protein as a key regulator of MUTE genes. The increase in the number of stomata on the adaxial leaf surface was observed under drought conditions. Stomata density and morphology had no significant correlation with the formation of wheat drought resistance. Parameters of stomata apparatus under drought conditions closely correlated with the width of the stoma on the abaxial side of the leaf. The data obtained are the basis for the creation of a fundamental model of the stomata biogenesis genes functioning in bread wheat. Product Description popup.authors Zborivska Oksana V.
