Objective: The purpose of the proposed study is to develop a model that includes the cocultivation of tumor and stromal cells to evaluate the migration potential of tumor cells under the influence of anticancer agents. Methods: cell culture, evaluation of locomotor properties of cells, immunochemical analysis, immunofluorescence analysis, image analysis. Not only the activity of transformed cells but also their environment - stromal cells is important for the migration activity of malignant tumor cells. Cancer-associated fibroblasts (CAFs) are considered to be one of the most common types of stromal cells that have an effect on cancer cells. They exert their regulatory influence through many signaling stages, including PI3K/AKT/ mTOR-dependent. During the fifth phase of the proposed project, the migratory potential of tumor cells was evaluated in the presence and absence of direct contact with fibroblasts and in the presence of an antitumor factor, namely the mTOR / S6K inhibitor of the rapamycin signaling cascade. Thus, a significant modulating effect of direct contact of tumor cells and fibroblasts on the dynamics of stratification of multicellular spheroids derived from MCF-7 breast cancer cells in the presence of an antitumor agent was found and cocultured fibroblasts were completely eliminated by cell levels.
