Molecular genetic mechanisms of production of and resistance to beta-lactam, glycopeptide and and phosphoglycolipid families are in the focus of the work. Goal of the work: to reveal novel genes for production of and resistance to to beta-lactam, glycopeptide and and phosphoglycolipid antibiotics. In the genomes of S. ghanaensis and A. teichomyceticus there were described for the first time the networks of global regulators of biosynthesis of moenomycins and teicoplanins. Mutations in genes for peptidoglycan glycosyltransferases were the only described type of mutational resistance to moenomycins prior to our work. Our results expand this model with the description of several novel resistance mechanisms. Properties of all putative teicoplanin resistance genes within teicoplanin biosynthesis gene cluster have been described for the first time. We developed new tools for systematic investigation of mechanisms of translational regulation, and applied them to AIPS. The obtained results support the theory of this project that it is possible to discover novel compounds and knowledge of antibiotic resistance mechanisms via genomic studies of AIPS producers.
