The aim of the work is to develop a test system based on the combination of techniques of long-term glycemic control molecular markers determination and assessment of oxidative-nitrative stress development, as well as the functional status of leukocytes – cells with decisive effect on the development of diabetic complications, and its correction by agmatine . Methods: biochemical, molecular biology, light microscopy, fluorescence microscopy, spectrophotometry. Results. It was shown a decrease of blood glucose concentration and glycosylated hemoglobin content in erythrocytes when administered agmatine to rats with diabetes. The amine caused a decrease in the content of advanced glycation end products (AGEs) in plasma and an increase in leukocytes in diabetic and control animals. Agmatine caused a restoration of the physiological levels of reactive oxygen species, reduction of the content of products of proteins oxidative modification, lipid peroxidation and 3-nitrotyrosine, which were elevated in leukocytes under diabetes. In addition, the activity of superoxide dismutase, catalase and glutathione peroxidase increased, and the activity of glutathione reductase was normalized. Under diabetes and agmatine administration, the level of polymerized actin in leukocytes decreased compared to pathology data, and it was found a quantitative redistribution of actin fractions (tightly associated with the cytoskeleton filaments, short actin filaments, and actin monomers). Thus, it is proved that agmatine is a powerful hypoglycemic, antioxidant and immunomodulatory agent. The polyamine restores the morpho-functional state of preactivated leukocytes under diabetes, improving a nonspecific link of immune protection and can be used as an effective drug.
